KCNK5蛋白在肺腺癌中的表达及临床意义

摘要:

目的: 旨在探究KCNK5蛋白在肺腺癌(LUAD)组织中的表达水平及其与临床病理特征、预后之间的关系。方法: 收集70例LUAD患者的癌组织及其癌旁组织(距肿瘤边缘5 cm处)标本,通过对LUAD组织的生物学信息研究及免疫组织方法检验LUAD组织及癌旁正常组织中KCNK5蛋白表达,分析其与LUAD临床病理特征的关系,采用单因素和多因素COX回归法分析影响LUAD患者预后的因素。结果: 免疫组化结果显示,KCNK5蛋白表达于LUAD细胞质,癌旁正常组织呈阴性或弱阳性;生物学信息分析得出LUAD组织KCNK5 mRNA表达均高于正常肺组织(P<0.05)。KCNK5蛋白高表达与LUAD患者的临床分期、淋巴结转移、分化程度有关联(P<0.05);多因素Logistic分析显示KCNK5蛋白高表达与淋巴结转移是影响肺腺癌预后的危险因素(P<0.05)。结论: KCNK5蛋白与LUAD患者淋巴结转移和预后密切相关,KCNK5蛋白有望成为肺腺癌早期诊断标志物及评估预后的指标。

Objective: To investigate the expression level of KCNK5 protein in lung adenocarcinoma(LUAD) tissues and the association between clinicopathological features and patient prognosis. Methods: Cancer tissue specimens from 70 LUAD patients and their paracancerous tissues(5 cm from the tumor margin) were collected. The biological information of LUAD tissues and the expression of KCNK5 protein in LUAD tissues and normal tissues adjacent to the cancer were examined by immunohistochemistry to analyze the connection between KCNK5 protein expression and the clinicopathological characteristics of LUAD, and the prognostic factors of LUAD were analyzed by binary and multiple regression methods. Results: The immunohistochemical results showed that KCNK5 protein was expressed in the cytoplasm of LUAD cells, and the normal tissues next to the cancer were negative or weakly positive; biological information analysis showed that the expression of KCNK5 mRNA in LUAD tissues was higher than that in the normal lung tissues(P<0.05); the expression of KCNK5 protein was closely correlated with the clinical stage, lymph node metastasis, and the degree of differentiation of the patients with LUAD(P<0.05); multifactorial logistic regression was used to analyze its relation with the clinicopathological features of patients with LUAD. Multifactorial Logistic regression analysis further confirmed that high KCNK5 protein expression and lymph node metastasis were independent risk factors affecting the prognosis of lung adenocarcinoma(P<0.05). Conclusion: KCNK5 protein is closely related to lymph node metastasis and prognosis of LUAD patients, and KCNK5 is expected to be an early diagnostic marker and prognostic indicator.
