lncRNA HAND2-AS1和GLUT-1在口腔鳞癌中的表达及其意义

摘要:

目的:检测口腔鳞癌患者中长链非编码RNA HAND2-AS1(lncRNA HAND2-AS1)和葡萄糖转运蛋白-1(GLUT-1)的表达情况,并探究两者与患者病理特征及预后的关系。方法:选取本院口腔科经病理检查确诊为口腔鳞癌患者68例,取其手术切除的口腔鳞癌组织和癌旁正常组织,采用实时荧光定量PCR法(qRT-PCR)分别检测癌组织及癌旁正常组织lncRNA HAND2-AS1与GLUT-1 mRNA表达水平,免疫组织化学法检测GLUT-1蛋白表达情况;通过Kaplan-Meier法绘制生存曲线评估lncRNA HAND2-AS1、GLUT-1蛋白表达对口腔鳞癌患者预后的影响;采用Cox回归分析口腔鳞癌患者预后的影响因素。结果:口腔鳞癌组织lncRNA HAND2-AS1表达水平低于癌旁正常组织,GLUT-1 mRNA表达水平、GLUT-1蛋白阳性表达率高于癌旁正常组织,差异有统计学意义(P<0.05)。lncRNA HAND2-AS1低表达组术后5年总生存率低于高表达组,GLUT-1蛋白阳性组术后5年总生存率低于阴性组,差异有统计学意义(P<0.05)。lncRNA HAND2-AS1、GLUT-1蛋白是影响口腔鳞癌患者预后的独立危险因素。结论:lncRNA HAND2-AS1在口腔鳞癌患者癌组织中的表达水平明显降低,GLUT-1 mRNA表达水平及GLUT-1蛋白阳性表达率明显升高,两者与患者临床病理特征及预后密切相关。

Objective:To detect the expression of long non-coding RNA HAND2-AS1(lncRNA HAND2-AS1) and glucose transporter 1(GLUT-1) in patients with oral squamous cell carcinoma, and to explore therelationship with pathological characteristics and prognosis of patients. Methods: A total of 68 patients with oral squamous cell carcinoma confirmed by pathological examination in the department of stomatology of our hospital were selected. The oral squamous cell carcinoma tissues and adjacent normal tissues resected during the surgery were taken. The expression level of lncRNA HAND2-AS1 and GLUT-1 mRNA in adjacent tissues and cancer tissues were detected with real-time fluorescent quantitative PCR(qRT-PCR), and the expression of GLUT-1 protein was detected with immunohistochemical method. The influence of lncRNA HAND2-AS1 and GLUT-1 protein expression on the prognosis of patients with oral squamous cell carcinoma was evaluated by drawing a survival curve with Kaplan-Meier method; and Cox regression analysis was used to analyze the prognostic factors of patients with oral squamous cell carcinoma. Results: The expression level of lncRNA HAND2-AS1 in oral squamous cell carcinoma group was lower than that in adjacent normal group, and the expression level of GLUT-1 mRNA and the positive expression rate of GLUT-1 protein were higher than those in the adjacent normal group, the difference was statistically significant(P<0.05). The 5-year overall survival rate of lncRNA HAND2-AS1 low expression group was lower than that of high expression group, and the 5-year overall survival rate of GLUT-1 protein positive group was lower than that of negative group, the difference was statistically significant(P<0.05). lncRNA HAND2-AS1 and GLUT-1 protein were independent risk factors affecting the prognosis of patients with oral squamous cell carcinoma. Conclusion: The expression level of lncRNA HAND2-AS1 in patients with oral squamous cell carcinoma is significantly decreased, and the expression level of GLUT-1 mRNA and the positive expression rate of GLUT-1 protein is significantly increased, they are closely related to the clinicopathological characteristics and prognosis of patients.
