血清外泌体microRNAs联合检测在食管癌早期诊断中的价值

摘要:

目的:探讨microRNA-21(miR-21)和microRNA-144(miR-144)在食管癌患者血清外泌体中的表达水平及其对食管癌的诊断价值。方法:选取2020年3月至2021年3月在我院就诊且经病理检查确诊的食管癌患者34例为食管癌组,选取同期体检健康者38例为对照组。分离并鉴定血清外泌体,提取血清外泌体miRNA。采用实时定量聚合酶链反应(qRT-PCR)检测miR-21和miR-144在两组血清外泌体中的表达水平。采用受试者工作特征(ROC)曲线评估血清外泌体中miR-21和miR-144对食管癌的诊断价值。结果:纯化的外泌体具有外泌体的典型特征。食管癌组血清外泌体miR-21表达水平高于对照组(P<0.05),miR-144表达水平低于对照组(P<0.05)。ROC曲线分析显示,miR-21和miR-144联合检测对食管癌诊断的准确度高于单独检测,差异均有统计学意义(P<0.05)。结论:血清外泌体miR-21和miR-144表达水平联合检测对诊断食管癌有潜在的应用价值。

Objective:To investigate the expression level and diagnostic value of microRNA-21(miR-21) and microRNA-144(miR-144) in serum exosomes of patients with esophageal cancer.Methods:A total of 34 patients with esophageal cancer diagnosed by pathological examination in Guangzhou Red Cross Hospital from March 2020 to March 2021 were selected as the esophageal cancer group,and 38 healthy people were selected as the control group.Serum exosomes were isolated and identified,and serum exosome miRNAs were extracted.Real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) was used to detect the expression levels of miR-21 and miR-144 in serum exosomes of the two groups.Receiver operating characteristic (ROC) curve was used to evaluate the diagnostic value of miR-21 and miR-144 in serum exosomes for esophageal cancer.Results:The purified exosomes had the typical characteristics of exosomes.The expression level of miR-21 in serum exosomes of esophageal cancer group was higher than that of control group (P<0.05).The expression level of miR-144 in esophageal cancer group was lower than that of control group (P<0.05).ROC curve analysis showed that the accuracy of the combined detection of miR-21 and miR-144 in the diagnosis of esophageal cancer was higher than that of the single detection,and the differences were statistically significant (P<0.05).Conclusion:The combined detection of miR-21 and miR-144 expression levels in serum exosomes has potential application value in the diagnosis of esophageal cancer.
