变应性鼻炎患者miR-21表达与Th1/Th2平衡的关系

摘要:

目的:检测变应性鼻炎(AR)患者血清微小RNA-21(miR-21)水平,并探讨其与Th1/Th2平衡的关系。方法:选取2017年1月至2019年7月于延安大学附属医院确诊的AR患者80例为实验组,另同期选取健康体检者85例为对照组。比较患者性别、年龄、身高、体质量指数(BMI)一般资料;采用实时荧光定量聚合酶链反应(qRT-PCR)测定血清miR-21表达水平;采用流式细胞术检测外周血单核细胞中Th1(%)、Th2(%)并计算Th1/Th2值;采用酶联免疫吸附法(ELISA)检测Th1细胞因子干扰素γ(IFN-γ)、白介素-12(IL-12)及Th2细胞因子白介素-4(IL-4)、白介素-13(IL-13)水平;Pearson相关性分析血清miR-21与Th1(%)、Th2(%)、Th1/Th2值及IFN-γ、IL-12、IL-4、IL-13水平相关性。结果:实验组患者血清miR-21、Th2(%)、Th2细胞因子IL-4、IL-13水平显著高于对照组(P<0.05),Th1(%)、Th1/Th2、Th1细胞因子IFN-γ、IL-12水平显著低于对照组(P<0.05)。Pearson相关性结果显示,AR患者血清循环miR-21水平与Th1(%)、Th1/Th2值、IFN-γ、IL-12水平均呈负相关(r=-0.411、-0.603、-0.623、-0.587,均P<0.05),与Th2(%)、IL-4、IL-13水平均呈正相关(r=0.501、0.457、0.529,均P<0.05)。结论:AR患者外周血CD4+T淋巴细胞Th1/Th2值严重失衡,且血清中miR-21异常高表达可能通过影响Th1/Th2平衡,参与AR病情进展过程,可为AR靶向防治提供一定思路。

Objective: To detect the serum microRNA-21 (miR-21) level in patients with allergic rhinitis (AR) and to explore its relationship with Th1/Th2 balance. Methods: A total of 80 patients with AR diagnosed in Affiliated Hospital of Yan'an Universityfrom January 2017 to July 2019 were selected as the experimental group, and another 85 healthy people were selected as control group at the same time. The general data of gender, age, height and body mass index (BMI) were compared; the expression level of miR-21 in serum was measured by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR); Th1 (%) and Th2 (%) in peripheral blood mononuclear cells were detected by flow cytometry, and the ratio of Th1/Th2 was calculated; The levels of Th1 cytokine interferon-γ (IFN-γ), interleukin-12 (IL-12), Th2 cytokines interleukin-4 (IL-4), interleukin-13 (IL-13) were detected by enzyme-linked immunosorbent assay (ELISA); In addition, Pearson correlation was used to analyze the correlation between serum miR-21 and Th1 (%), Th2 (%), Th1/Th2, IFN-γ, IL-12, IL-4, IL-13 levels. Results: The levels of serum miR-21, Th2 (%), Th2 cytokines IL-4 and IL-13 in the experimental group were significantly higher than those in control group (P<0.05), while the levels of Th1 (%), Th1/Th2, Th1 cytokine IFN-γ and IL-12 were significantly lower than those in the control group (P<0.05). Pearson correlation results showed that the level of serum miR-21 was negatively correlated with Th1 (%), Th1/Th2 ratio, IFN-γ, IL-12 (r=-0.411,-0.603,-0.623,-0.587, all P<0.05), and positively correlated with Th2 (%), IL-4, IL-13 (r=0.501, 0.457, 0.529, all P<0.05). Conclusion: Th1/Th2ratio in CD4+ T lymphocytes in peripheral blood of AR patients is seriously unbalanced, and the abnormal high expression of miR-21 in serum may affect the balance of Th1/Th2 and participate in the progression of AR, which may provide some ideas for the targeted prevention and treatment of AR.
