鼻咽癌中miR-221-3p的表达及临床意义

摘要:

目的: 检测鼻咽癌中微小RNA-221-3p (miR-221-3p)的表达,分析其与增殖、凋亡指标的相关性。方法: 选取鼻咽癌患者69例作为观察组,慢性鼻炎患者69例作为对照组。均留取组织标本,应用实时荧光定量聚合酶链反应法检测两组中miR-221-3p的表达。蛋白免疫印迹法检测鼻咽癌中BCL-2相关X蛋白(BAX)的表达。免疫组织化学法检测鼻咽癌中Ki67的表达。原位杂交法检测鼻咽癌中EB病毒的表达。结果: 观察组miR-221-3p表达高于对照组,差异有统计学意义(P<0.05),miR-221-3p的表达在观察组不同组织学亚型、淋巴结转移、局部浸润程度的分组中差异有统计学意义(P<0.05)。生存分析显示miR-221-3p高表达患者的生存时间短(P<0.05)。Spearman相关分析显示,miR-221-3p表达与Ki67增殖指数呈正相关(r=0.56,P<0.05),与BAX呈负相关(r=-0.69,P<0.05)。结论: 与慢性鼻炎患者比较,鼻咽癌中miR-221-3p表达升高。MiR-221-3p高表达促进肿瘤增殖、抑制凋亡,预示病变呈进展状态及较差的预后。

Objective: To detect the expression of microRNA-221-3p(miR-221-3p) in nasopharyngeal carcinoma, analyze its correlation with cell proliferation and apoptosis indexes. Methods: A total of 69 patients with nasopharyngeal carcinoma were selected as the observation group. 69 cases of chronic rhinitis were selected as the control group. Tissue sample were retained. Expression of miR-221-3p was detected by real-time fluorescent quantitative polymerase chain reaction(qPCR) in two groups. Expression of BCL2 associated X protein(BAX) was detected by Western blotting in observation group. Expression of Ki67 was detected by immunohistochemistry in observation group. Expression of EBV was detected by in situ hybridization in observation group. Results: Expression of miR-221-3p in observation group was significantly higher than that in control group (P<0.05). Expression of miR-221-3p was significant different in histological subtypes, lymph node metastasis and local infiltration degree (all P<0.05). MiR-221-3p was negatively related to survival time by survival analyze(P<0.05). Positive correlation was found between miR-221-3p and Ki67 proliferation index (r=0.56, P<0.05). Negative correlation was found between miR-221-3p and BAX(r=-0.69, P<0.05). Conclusion: Compared with the control group, expression of miR-221-3p was higher in observation group. The high expression of miR-221-3p promotes tumor proliferation and inhibits apoptosis, which indicates the poor biological behavior and prognosis.
