miR142-5p、miR144-3p在甲状腺乳头状癌中的功能研究

摘要:

目的: 研究miR142-5p、miR144-3p在甲状腺乳头状癌中表达情况及其功能。方法: 实时荧光定量聚合酶链反应(qRT-PCR)检测60例不同TNM分期的甲状腺乳头状癌及癌旁组织中的miR142-5p、miR144-3p表达水平;流式细胞术检测miR142-5p、miR144-3p对甲状腺乳头状癌细胞(B-CPAP细胞)凋亡的影响。结果: 甲状腺乳头状癌组织中miR142-5p和miR144-3p的表达水平低于癌旁组织,差异均有统计学意义(均P<0.01),随着TNM分期增加两者表达水平降低,差异均有统计学意义(均P<0.01);流式细胞术检测结果显示miR142-5p及miR144-3p实验组凋亡率分别为(19.11±5.80)%、(12.72±1.13)%,对照组的凋亡率为(7.22±0.71)%,空载组凋亡率为(4.29±0.61)%,实验组细胞凋亡率高于对照组及空载组,差异均有统计学意义(均P<0.05)。结论: miR142-5p、miR144-3p在甲状腺乳头状癌细胞内表达低于癌旁细胞内表达,表达水平随肿瘤分期增加而降低,其可能在甲状腺乳头状癌的发生发展中起到抑癌作用。

Objective: To investigate the expression and function of miR142-5p and miR144-3p in thyroid papillary carcinoma.Methods: Real-time fluorescence quantitative polymerase chain reaction(qRT-PCR) was used to detect the expression of miR142-5p and miR144-3p in thyroid papillary carcinoma and paracancerous tissues of 60 cases with different TNM stages. Flow cytometry was used to detect the effect of miR142-5p and miR144-3p on the apoptosis of thyroid papillary carcinoma cells(B-CPAP).Results: The expression of miR142-5p and miR144-3p in thyroid papillary carcinoma tissues were lower than those in paracancerous tissues, and the differences were statistically significant(all P<0.01). With TNM stage increased,the expression of miR142-5p and miR144-3p in thyroid papillary carcinoma tissues decreased, and the differences were statistically significant (all P<0.01).Flow cytometry detection results showed that the apoptosis rate of miR142-5p and miR144-3p groups was (19.11±5.80)% and (12.72±1.13)%, respectively; the apoptosis rate of the control group was (7.22±0.71)%, and that of the no-load group was (4.29±0.61)%. The apoptosis rate of the miR142-5p and miR144-3p group were higher than those of the control group and no-load group, the differences were statistically significant(all P<0.05).Conclusion: The expression of miR142-5p and miR144-3p in papillary thyroid carcinoma tissues were lower than those in paracancerous tissues, and they decreased with the increase of tumor TNM stage, which may play a role in inhibiting the occurrence and development of thyroid papillary carcinoma.
