LncRNA lncRNA PCGEM1和LncRNA lncRNA PVT1在哮喘患者血清中表达与意义

摘要:

目的:检测长链非编码RNA(lncRNA)前列腺癌基因表达标记物1(PCGEM1)和lncRNA细胞瘤多样异位基因1(PVT1)在哮喘(BA)患者血清中的表达水平,探讨其临床意义。方法:前瞻性选取2016年1月—2019年4月经本院收治确诊的93例BA患者作为观察组,同期选取90例体检健康人群作为对照组;采用实时荧光定量PCR(qRT-PCR)法检测血清中lncRNA PCGEM1、lncRNA PVT1水平;酶联免疫吸附试验(ELISA)法测定血清中白细胞介素6(IL-6)、白细胞介素17(IL-17)、肿瘤坏死因子α(TNF-α)水平;Pearson法分析BA患者血清中lncRNA PCGEM1、lncRNA PVT1与IL-6、IL-17、TNF-α表达的相关性;利用受试者工作特性曲线(ROC)预测lncRNA PCGEM1、lncRNA PVT1表达对BA的诊断价值。结果:观察组血清中lncRNA PCGEM1表达水平显著低于对照组(P<0.05),lncRNA PVT1、IL-6、IL-17、TNF-α表达水平均显著高于对照组(P<0.05)。BA患者血清中lncRNA PCGEM1与IL-6、IL-17、TNF-α表达呈负相关(r=-0.551、-0.686、-0.602,P<0.05),lncRNA PVT1与IL-6、IL-17、TNF-α表达呈正相关(r=0.709、0.694、0.637,P<0.05);lncRNA PCGEM1与lncRNA PVT1表达呈负相关(r=-0.634,P<0.05)。lncRNA PCGEM1诊断BA的ROC曲线下面积为0.859,截断值为0.52,灵敏度为89.2%,特异性为70.0%;lncRNA PVT1诊断BA的ROC曲线下面积为0.892,截断值为2.36,灵敏度为82.8%,特异性为85.6%;两者联合诊断BA的ROC曲线下面积为0.949,截断值为0.47,灵敏度为90.3%,特异性为88.9%。结论:BA患者血清中lncRNA PCGEM1呈低表达,lncRNA PVT1呈高表达,两者呈负相关,对BA均有一定的诊断价值,且两者联合检测的诊断价值更高。

Objective: To detect the expression levels of prostate cancer gene expression marker 1 (PCGEM1) and lncRNA plasmacytoma variant translocation 1 (pvt1) in the serum of patients with asthma (BA) and to explore their clinical significance.Methods: From January 2016 to April 2019, 93 patients with BA diagnosed in our hospital were prospectively selected as the observation group and 90 healthy people as the control group; the levels of lncRNA PCGEM1 and lncRNA PVT1 in serum were detected by real-time fluorescence quantitative PCR (qRT-PCR); the levels of interleukin-6 (IL-6), interleukin-17 (IL-17) and tumor necrosis factor-α (TNF-α) in serum were measured by enzyme-linked immunosorbent assay (ELISA). Pearson method was used to analyze the correlations between the expressions of lncRNA PCGEM1, lncRNA PVT1, IL-6, IL-17 and TNF-α in serum of BA patients; receiver operator characteristic curves (ROC) was used to predict the diagnostic values of lncRNA PCGEM1 and lncRNA PVT1 expressions for BA.Results: The expression level of lncRNA PCGEM1 in serum of the observation group was significantly lower than that of the control group (P<0.05), and the expression levels of lncRNA PVT1, IL-6, IL-17 and TNF-α were significantly higher than those in the control group (P<0.05). There was a negative correlation between the expression of lncRNA PCGEM1 and IL-6, IL-17, TNF-α (r =-0.551,-0.686,-0.602, P<0.05), lncRNA PVT1 was positively correlated with the expressions of IL-6, IL-17 and TNF-α (r=0.709, 0.694, 0.637, P<0.05); and there was a negative correlation between pcgem1 and pvt1 (r=-0.634, P<0.05). The area under ROC curve of lncRNA PCGEM1 in BA diagnosis was 0.859, the cutoff value was 0.52, the sensitivity was 89.2%, and the specificity was 70.0%; the area under ROC curve of lncRNA PVT1 in BA diagnosis was 0.892, the truncation value was 2.36, the sensitivity was 82.8%, and the specificity was 85.6%; the area under ROC curve of combined diagnosis in BA diagnosis was 0.949, the cutoff value was 0.47, the sensitivity was 90.3%, and the specificity was 88.9%.Conclusion: The serum lncRNA PCGEM1 of BA patients shows low expression and high lncRNA PVT1 expression, which were negatively correlated and had certain diagnostic value for BA, and the diagnostic value of combined detection was higher.
