LncRNA-GAS5在食管鳞癌中的表达及其对细胞增殖和凋亡的影响

摘要:

目的:探讨长链非编码核糖核酸生长组织特异性转录体5(LncRNA-GAS5)在食管鳞癌组织中的表达及其对食管鳞癌细胞株EC9706细胞增殖、凋亡的影响。方法:选取2016年2月至2019年1月于中国人民解放军陆军第958医院诊治的食管鳞癌患者78 例,比较LncRNA-GAS5在食管鳞癌组织及癌旁组织中的表达差异;过表达质粒(pEGFP-C1-GAS5)转染EC9706后通过CCK-8试剂盒以及平板克隆试验检测LncRNA-GAS5过表达对细胞增殖能力的影响,采用Annexin V/PI双染流式细胞术检测LncRNA-GAS5过表达对细胞凋亡的影响,荧光实时定量聚合酶链反应(qRT-PCR)及蛋白质免疫印迹法检测抑癌基因PTEN表达变化。结果:与癌旁组相比,食管鳞癌组LncRNA-GAS5相对表达量降低(P<0.05);与空白组、阴性对照组相比,pEGFP-C1-GAS5组LncRNA-GAS5相对表达量升高(P<0.05);与空白组、阴性对照组相比,pEGFP-C1-GAS5组转染后48、72、96 h OD值、细胞克隆数降低(P<0.05),pEGFP-C1-GAS5组细胞凋亡率升高(P<0.05),PTEN mRNA、PTEN蛋白表达升高(P<0.05)。结论:食管鳞癌患者癌组织内LncRNA-GAS5低表达,LncRNA-GAS5过表达抑制癌细胞增殖,促进癌细胞凋亡,可能是通过促进抑癌基因PTEN表达实现的。

Objective: To investigate the expression of long non-coding ribonucleic acid growth tissue-specific transcript 5 (LncRNA-GAS5) in esophageal squamous cell carcinoma and its effects on the proliferation and apoptosis of EC9706 cells. Methods: 78 patients with esophageal squamous cell carcinoma were selected from February 2016 to January 2019 in 958 Hospital of the Chinese People's Liberation Army, the expression of LncRNA-GAS5 in esophageal squamous cell carcinoma and adjacent tissues was compared; after transfection of EC9706 with pEGFP-C1-GAS5, the effect of over expression of LncRNA-GAS5 on cell proliferation was detected by CCK-8 kit and plate cloning assay, Annexin V/PI double staining flow cytometry was used to detect the effect of overexpression of LncRNA-GAS5 on cell apoptosis, the expression of PTEN was detected by quantitative real time polymerase chain reaction(qRT-PCR) and western blotting. Results: Compared with the adjacent group, the relative expression of LncRNA-GAS5 in esophageal squamous cell carcinoma group was lower (P<0.05); compared with blank group and negative control group, the relative expression of LncRNA-GAS5 in pEGFP-C1-GAS5 group increased (P<0.05); compared with blank group and negative control group, OD value and cell clone number in pEGFP-C1-GAS5 group decreased at 48, 72, 96 hours after transfection (P<0.05), the apoptotic rate of pEGFP-C1-GAS5 group was increased (P<0.05), the expressions of PTEN mRNA and PTEN protein increased (P<0.05). Conclusion: The expression of LncRNA-GAS5 in esophageal squamous cell carcinoma is low, LncRNA-GAS5 overexpression inhibits the proliferation of cancer cells and promotes the apoptosis of cancer cells, probably by promoting the expression of tumor suppressor gene PTEN.
