lncRNA NEAT1在急性髓系白血病患者外周血中的表达及其与预后的关系

摘要:

目的:探究长链非编码RNA NEAT1(lncRNA NEAT1)在急性髓系白血病(AML)患者外周血中的表达,分析其与临床预后的关系。方法:选择2011年3月至2014年3月本院收治的AML患者76例(AML组)和来本院体检的健康人32例(对照组)作为研究对象,采集各个研究对象的外周血液标本,采用实时荧光定量PCR(qRT-PCR)法检测外周血单个核细胞中lncRNA NEAT1的表达;分析lncRNA NEAT1表达与AML患者临床病理特征的关系;对出院后AML患者进行随访,分析lncRNA NEAT1表达与AML患者预后的关系。结果:lncRNA NEAT1在AML患者外周血单个核细胞中表达明显高于对照组(P<0.05);lncRNA NEAT1在AML患者外周血单个核细胞中的表达与患者的年龄、性别、FAB亚型无关(P>0.05),与危险度分层、白细胞(WBC)含量、血红蛋白(Hb)含量及血小板(PLT)含量有关(P<0.05);lncRNA NEAT1高表达组AML患者5年生存率为32.50%,显著低于低表达组的55.56%(P<0.05);多因素分析显示,lncRNA NEAT1表达和危险度分层是影响AML患者预后的独立危险因素(HR=2.513,95% CI 1.327~4.759;HR=2.781,95% CI 1.753~4.412,均P<0.05)。结论:lncRNA NEAT1在AML患者外周血单个核细胞中高表达,其表达变化可以反映AML的发生、发展进程,可能作为AML预后判断的一个重要参考指标。

Objective: To investigate the expression of long non-coding small RNA NEAT1 (lncRNA NEAT1) in peripheral blood of acute myeloid leukemia (AML) and to analyze its relationship with clinical prognosis. Methods: 76 AML patients (AML group) admitted to our hospital and 32 healthy persons (control group) who came to our hospital for physical examination from March 2011 to March 2014 were collected as the research objects. Peripheral blood samples were collected from all subjects, the expression of lncRNA NEAT1 in peripheral blood mononuclear cells was detected by real-time fluorescence quantitative PCR (qRT-PCR); the relationship between NEAT1 expression with clinicopathological features of AML patients was analyzed; after discharge, AML patients were followed-up, the relationship between the expression of lncRNA NEAT1 with the prognosis of AML patients was analyzed.Results: The expression of lncRNA NEAT1 in peripheral blood mononuclear cells of AML patients was significantly higher than that of control group (P<0.05); the expression of lncRNA NEAT1 in peripheral blood mononuclear cells of AML patients was not related to age, sex and FAB subtype (P> 0.05), but related to risk stratification, white blood cell (WBC) content, hemoglobin(Hb) content and platelet(PLT) content (P< 0.05); the 5-year survival rate of patients with AML in the lncRNA NEAT1 high expression group was 32.50%, which was significantly lower than 55.56% in the low expression group (P<0.05). Multivariate analysis showed that the expression of lncRNA NEAT1 and risk stratification were independent risk factors affecting the prognosis of AML patients (HR=2.513, 95% CI 1.327-4.759; HR=2.781, 95% CI 1.753-4.412, all P<0.05). Conclusion: LncRNA NEAT1 is highly expressed in peripheral blood mononuclear cells of AML patients, and its expression change can reflect the occurrence and development of AML, which may be an important prognostic indicator for AML.
