Cyr61及CTGF的表达水平与肺癌进展的相关性分析

摘要:

目的:探讨富含半胱氨酸61(Cyr61)及结缔组织生长因子(CTGF)表达水平的变化与肺癌进展的关系。方法:选取2014年1月至2017年10月石家庄市第一医院接诊的102例肺癌患者为研究对象,将肺癌患者手术后的癌组织设为研究组,对应的癌旁组织(距离癌组织5 cm)为对照组。采用ELISA法检测Cyr61和CTGF mRNA的表达水平,采用免疫组化法测定Cyr61和CTGF蛋白表达情况。结果:Cyr61mRNA表达水平及其蛋白表达阳性率研究组低于对照组,CTGF mRNA表达水平及其蛋白表达阳性率研究组高于对照组,差异有均有统计学意义(P<0.05)。Cyr61与CTGF mRNA的表达水平呈正相关(r=0.612,P=0.004);Cyr61和CTGF蛋白表达在不同病理类型、病理分期以及是否吸烟、有无家族史、淋巴结是否转移患者之间差异有统计学意义,CTGF蛋白的表达在不同位置的肿瘤之间差异有统计学意义(P<0.05)。研究组组织中72%的Cyr61表达可由病理分型、淋巴结转移、吸烟等8个自变量解释,62%的CTGF表达可由肿瘤位置、吸烟、性别等7个自变量解释。结论:Cyr61与CTGF表达水平与患者临床病理参数密切相关,能够有效反映肺癌细胞的侵袭转移能力和恶性程度。

Objective:To investigate the correctation between the expression of cysteine-rich 61(Cyr61),connective tissue growth factor (CTGF) and the progression of lung cancer.Methods:One hundred and two patients with lung cancer admitted to First Hospital of Shijiazhuang from January 2014 to October 2017 were selected as the study subjects. The cancer tissues were set as the study group, and the corresponding adjacent tissues (5 cm away from the cancer tissue) were used as the control group. The expression levels of Cyr61 and CTGF mRNA were detected by ELISA. The expression of Cyr61 and CTGF protein was determined by immunohistochemistry. Results:The expression level of Cyr61 mRNA and the positive rate of protein expression were lower in the study group than those in the control group. The expression level of CTGF gene mRNA and the positive rate of protein expression were higher in the study group than those in the control group, and the difference were statistically significant (all P<0.05). There was a positive correlation between Cyr61 and CTGF mRNA (r=0.612, P=0.004). There was a statistically significant difference between Cyr61 and CTGF protein expression in patients with different pathological types, pathological stages, smoking, family history, and lymph node metastasis. The difference of CTGF protein expression in different tumor locations was statistically significant (P<0.05).Seventy-two percent of Cyr61 expression in the study group could be explained by 8 independent variables such as pathological type, lymph node metastasis and smoking, and so on. Sixty-two percent of CTGF expression could be explained by 7 independent variables such as tumor location, smoking and gender, and so on.Conclusion:Cyr61 and CTGFexpression are closely related to clinical pathological parameters of patients, and can effectively reflect the invasion and metastasis ability and malignant degree of lung cancer cells.
