间苯三酚联合运动对原发性痛经大鼠血清学指标与子宫组织MMP-3与MMP-9表达的影响

摘要:

目的:探讨间苯三酚联合运动对原发性痛经大鼠血清学指标与子宫组织MMP-3与MMP-9表达的影响。方法:4月龄未孕动情间期SD雌性大鼠24只,随即分为对照组、模型组和治疗组(注射间苯三酚+运动干预),利用缩宫素建立原发性痛经模型。比较各组大鼠扭体情况,测定各组大鼠血清血清雌二醇(E2)、前列腺素E2(PGE2)、前列腺素F2α(PGF2α)、雌激素(P)的含量,Real-time PCR和Western blot方法检测分别检测子宫组织MMP-3、MMP-9基因及蛋白表达。结果:间苯三酚联合运动干预,能够显著上调原发性痛经大鼠血清PGE2水平,降低血清E2、PGF2α水平比值,降低扭体反应次数,P<0.05。与对照组相比,原发性痛经大鼠子宫组织MMP-3、MMP-9基因及蛋白表达水平显著升高,P<0.05;但是给予间苯三酚联合运动干预后,原发性痛经大鼠子宫组织MMP-3、MMP-9基因及蛋白表达水平显著降低,P<0.05。结论:间苯三酚联合运动可拮抗缩宫素致大鼠实验性痛经。

Objective:To investigate the effects of phloroglucin combined with training on serological indicators and expression of MMP-3 and MMP-9 in uterus of primary dysmenorrhea rats.Methods:Twenty-four female SD rats were randomly divided into three groups:control group, model group and treatment group. Oxytocin was used to establish primary dysmenorrhea model. The levels of estradiol (E 2), prostaglandin E 2 (PGE2), prostaglandin F 2 α (PGF2 α) and estrogen (P) were measured. The expression of MMP-3,MMP-9 gene and protein were detected by Real-time PCR and Western blot.Results:In combination with exercise intervention, the level of serum PGE2 in primary dysmenorrhea rats was upregulated, the ratio of serum E2PGF2 α was decreased, and the times of writhing reaction was decreased (P<0.05). Compared with the control group, the expression of MMP-3,MMP-9 gene and protein in uterus of primary dysmenorrhea rats was significantly increased (P<0.05). However, the expression of MMP-3,MMP-9 gene and protein in uterine tissues of primary dysmenorrhea rats decreased significantly (P<0.05) after treatment with phloroglucinol combined with exercise. Conclusion:Phloroglucinol combined with exercise can antagonize experimental dysmenorrhea induced by oxytocin in rats.
