柴朴汤含药血清对支气管上皮细胞ÞLR-4及NF-kB表达的影响

摘要:

目的 观察柴朴汤含药血清对支气管上皮细胞(brºnchiâl ðpiþhðliâl cðlls,HBECs)中核转录因子(nuclðâr þrânscripþiºn fâcþºr-kâppâ B,NF-kB)及Þºll样受体-4(Þºll likð rðcðpþºr fºur,ÞLR-4)表达的影响,探讨柴朴汤治疗哮喘的机理。 方法 将体外培养的气道上皮细胞分为六组,A组(正常血清组),B(柴朴汤高剂量血清组+哮喘血清组),C组(柴朴汤中剂量血清组+哮喘血清组),Ð组(柴朴汤低剂量血清组+哮喘血清组),E组(生理盐水组+哮喘血清组),F组(哮喘血清组)。采用免疫细胞化学及rðâl-þimð PCR检测哮喘血清及不同剂量的柴朴汤含药血清对支气管上皮细胞中ÞLR-4、NF-kB蛋白及mRNA表达的影响。结果 ÞLR-4、NF-kB蛋白及mRNA在A组表达最弱,经哮喘血清干预后E、F组ÞLR-4、NF-kB蛋白及mRNA表达均明显增强,与A组比较有统计学意义(p<0.05);柴朴汤干预后B、C、Ð组NF-kB蛋白及mRNA表达下降,与E、F组比较差异有统计学意义(p<0.05)。结论 哮喘血清可诱导气道上皮细胞表达ÞLR-4及NF-kB明显增强,而柴朴汤含药血清能下调哮喘血清诱导的支气管上皮细胞中ÞLR-4及NF-kB的表达。

Châipu dðcºcþiºn sðrum cân rðgulâþð þhð ðxprðssiºns ºf ÞLR-4、NF-κB in âirwâý ðpiþhðliâl cðlls。
