TCF7L2基因与2型糖尿病发生相关性的研究进展

TCF7L2基因是Wnt信号通路的转录因子,表达于肠道、胰腺等多种组织。Wnt信号通路在胰岛β细胞的增殖及胰岛素的分泌中起重要作用,并影响小肠L细胞胰高血糖素样肽-1(GLP-1)的合成。TCF7L2基因是目前全基因组关联研究(GWAS)发现的与2型糖尿病(T2DM)关联度最高的基因之一,并在多种族研究中得以证实,其单核苷酸多态性及选择性剪接会影响胰岛β细胞功能及胰岛素抵抗。TCF7L2基因与T2DM相关性的深入研究与认识,为T2DM的干预和治疗提供新思路。

TCF7L2 is a Wnt signaling-associated transcription factor expressed in several tissues, including gut and pancreas. Wnt signaling plays an important role in β-cell proliferation and insulin secretion and influences synthesis of glucagon-like peptide-1 (GLP-1) in intestinal L-cells. The association between TCF7L2 gene variants and type 2 diabetes mellitus(T2DM) has been confirmed by the genome-wide association study (GWAS) and a lot of other studies in different ethnic groups. Single nucleotide polymorphisms and alternative splicing of TCF7L2 affect both the pancreatic β cell function and insulin resistance. Further study will provide new pathways for the prevention and treatment of T2DM.
