FK506对d-BSA超载的NRK-52E细胞增殖的影响

摘要:

目的:探讨他克莫司(tacrolimus,FK506)对去脂牛血清白蛋白(denatured bovine serum albumin, d-BSA)超载的大鼠肾小管上皮细胞(The normal rat kidney cell line,NRK-52E)增殖活性的影响。方法:将不同浓度FK506于不同时间作用于d-BSA超载的NRK-52E细胞,通过四甲基偶氮唑盐法(MTT)测定不同浓度FK506与d-BSA共孵育NRK-52E细胞的增殖活性。结果:①与对照组相比,终质量浓度为1mg/ml的d-BSA对NRK-52E细胞无明显促进增殖的作用,与d-BSA孵育时间无明显关系(P>0.05);作用6后,终质量浓度为5、10、20、50mg/ml的d-BSA能够促进NRK -52E细胞增殖(P<0.05)。②终质量浓度为0.1ng/ml的FK506预处理NRK-52E细胞4h后与不同浓度d-BSA共孵育引起的细胞增殖与对照组无异(P>0.05);终质量浓度为1、10、20ng/ml的FK506预处理4h后与20mg/ml的d-BSA共孵育6h,具有抑制d-BSA引起的细胞增殖的作用,并呈现与FK506浓度与时间的依赖关系(P<0.05)。结论:d-BSA超载NRK-52E可以引起细胞增殖;FK506具有抑制d-BSA促增殖的作用。

Objective:To investigate effect of FK506 on d-BSA-induced proliferation of NRK-52E cells. Methods:Different concentrations of FK506 on d-BSA-induced NRK-52E cells, the cells were observed at different times within proliferation.Proliferation was determined by MTT assay. Results:①While l mg/ml d-BSA did not have any significant difference on NRK-52E proliferation that was not related with time(P>0.05);5,10,20,50mg/ml d-BSA could induce proliferation of NRK-52E for 6 hours(P<0.05).②0.1ng/ml FK506 pretreatment with NRK-52E for 4 hours did have no significance on d-BSA-induced proliferation of NRK-52E cells(P>0.05);1,10,20ng/ml FK506 commonly incubated for 6 hours with d-BSA after pretreatment for 4 hours,proliferation was inhibited in a dose- and time- dependent manner(P<0.05). Conclusion:D-BSA may induce proliferation in NRK-52E that FK506 could inhibit.
