【Abstract】Objective To explore the different does of Ovalbumin(OVA) sensitization whether play a fluence in airway hyperresponsiveness of mice model of asthma. Methods The mice were randomly divided into 3 groups ,with 10 mice in each group: control group and asthmatic groups(including asthma A group ,the does OVA sensitization is 2 mg every time ; and asthma B group,which is 10 ug ). The asthma A、B group were sensitized in the 0th、14th day with different does of OVA each other ,then inhaled ovalbumin(OVA)beginning the 21st day ,and lasting for 6 days . to observe the breathing and the color of labiali and tail mucosa.The last time OVA stimulateed, after 12 hours ,the mice were measured the Penh to evaluate the airway responsiveness by Noninvasive lung functional instrument , bronchoalveolar lavage cytology was performed and lung tissue sections were prepared for histopathologic examination to evaluate the airway inflammation.examine the IgE、IFN-gama of blood and BALF by ELISA method. Results①the skin of mice lip and tail was caynosis after the second allergic sensitization in asthmatic B group mice.②The airway hyperrespomsiveness in asthmatic group was significantly higher than that in the control mice,and asthmatic B group`s Penh was significantly higher than asthmatic A group; ③The total cell number and the eosinophils in BALF of the asthmatic mice were significantly higher than that in the control mice ( both P< 0.05);The total cell number and the eosinophils in BALF of the asthmatic B group were higher than that in asthmatic A group , but p>0.05; ④the contents of IgE in peripheral blood and BALF of the asthmatic mice were significantly higer than that in the control mice, P<0.01,and he asthmatic B group (37.47±6.18,26.10±7.18)were higher than that in asthmatic A group (26.09±3.76,P<0.01;12.47±3.02,P<0.01).⑤ the contents of IFN-gama in peripheral blood and BALF of the asthmatic mice were lower than that in control ones, and the asthmati |
