Objective: Research on the effects of difference dose Ibandronate of fracture healing after local application and to elect the best appropriate dose. Methods: forty-five healthy New Zealand rabbits were randomly divided into five groups: 0ml, 0.1ml, 0.2ml, 0.4ml, 9 rabbits for each group. All the rabbits were made 3 mm wide bone defection at lower one third part of unilateral radius as fracture healing model.0ml group regard as the control group (CG) in the five groups and no more management after the model was built. Each of those was given different Ibandronate according to each group’s setting dose through dosage locally. At week 1, 2, 4 the state of fracture healing were examined with DR, and then scored. At week 1, 2, 4 three rabbits were selected from each group randomly and were killed by aeroembolism respectively. Their radial bone on side of model were anatomied abruption after examined with DR and test through biomechanics. Then the calluses were intercepted from fracture sites as the research objects. Pathological changes were observed in callus samples and Immunohistochemistry stain were performed to examine receptor of VEGF expression in tissues of callus. Results: ①Radiographic analysis: At week 2,4 the local application group obtained higher scores than in CG(p<0.05).The group of 0.3ml obtained higher scores than 0.1ml,0.2ml group(p<0.05)and compare for 0.4ml group was no contrast(p>0.05).②Biomechanics test Index: At week 4 anti-bend strength of the local application group obtained index higher than in CG(p<0.05).The group of 0.3ml index higher than 0.1ml,0.2ml group(p<0.05)and compare for 0.4ml group was no contrast(p>0.05); Max-offset of the local application obtained higher index than in CG(p<0.05),but compare in each of the local application groups was no contrast(p>0.05).③Osteoblastic index(OBI):At week 1,2,4, the local application group OBI higher than in CG(p<0.05).The group of 0.3ml OBI higher than 0.1ml,0.2ml group(p<0. |
