胶质瘤干细胞分离鉴定及其化疗敏感性研究

摘要:

目的 探讨脑肿瘤干细胞(brain tumor stem cells,BTSC)的分离、培养、鉴定,研究BTSC与胶质瘤在化疗敏感性的差异。方法 将三株人胶质瘤细胞SHG-44、U251、U87MG细胞分别接种于无血清培养基,从而分离出各自脑肿瘤干细胞;扫描电镜观察BTSC的形态;RT-PCR法检测干细胞CD133、GFAP的表达;采用CCK8法检测化疗药物顺铂(cisplatin,CDDP)及长春新碱(vincristine,VCR)作用后胶质瘤及BTSC细胞活性变化。结果 从人脑胶质瘤细胞分离出脑肿瘤干细胞BTSC,BTSC可在体外增殖及传代,表达特异性标志物CD133,不表达GFAP。CCK8法检测出BTSC较普通细胞耐药更为耐药。结论 脑肿瘤细胞中存在BTSC,且BTSC较普通胶质瘤细胞化疗敏感性降低。

Objective To explore the methods of isolation,culture and identification of brain tumor stem cells (BTSC),and to study the difference of chemosensitivity between the BTSC and the glioma cells. Methods Three kinds of glioma cells were respectly seeded into serum-free medium and then disassociated each BTSC, the 形态 can be observed under the scanning electron microscope. The expression of CD133/GFAP of BTSC was detected by RT-PCR, the CCK8 was used to evaluated the difference of proliferation between the glioma cells and BTSC both of which are used after CDDP and VCR. Results The brain stem cells isolated from glioma, have the ability to propagate and differationate in vitro.The character of expression CD133 was proved by flow cytometry. Conclusion BTSC is existed in glioma, and BTSC is more resistant to chemical therapy than glioma cell.
