甘草甜素对人舌癌Cal-27细胞增殖、凋亡及侵袭能力的影响

摘要:

目的: 研究甘草甜素对人舌癌Cal-27细胞生物学行为的影响。方法: 使用不同浓度的甘草甜素(10、20、40和80 μmol·L-1)处理人舌癌Cal-27细胞不同时间,分别采用细胞增殖实验(CCK-8法)检测细胞活力;流式细胞术检测细胞凋亡情况;转移小室实验检测细胞侵袭能力;蛋白质免疫印迹法检测细胞中Bcl-2、Bax及p-Capase-3的蛋白表达水平。结果: 甘草甜素对Cal-27细胞的增殖有较强的抑制作用(P<0.05),且呈时间和剂量依赖性,同时可显著诱导细胞凋亡(P<0.05),抑制细胞的侵袭能力(P<0.05)。蛋白质免疫印迹法检测结果显示,与对照组相比,甘草甜素处理后细胞中Bcl-2的蛋白表达水平明显降低(P<0.05),Bax及p-Capase-3的蛋白表达水平明显增加(P<0.05)。结论: 甘草甜素能够抑制人舌癌Cal-27细胞株的体外增殖及侵袭能力并促进细胞凋亡,其机制可能是通过上调Bax和p-Capase-3的表达、下调Bcl-2的表达实现的。

Objective: To investigate the effects of glycyrrhizin on the biological behavior of human tongue cancer Cal-27 cells.Methods: Cal-27 cells were treated with different concentrations of glycyrrhizin (10, 20, 40 and 80 μmol·L-1) for different time. Cell viability was detected by CCK-8 method. Apoptosis was detected by flow cytometry. The cell invasion ability was detected by Transwell assay. Western blot was used to detect the protein expression levels of Bcl-2, Bax and p-Capase-3 in cells. Results: Glycyrrhizin had a strong inhibitory effect on the proliferation of Cal-27 cells (P<0.05) in a time-and dose-dependent manner. Glycyrrhizin treatments also significantly induced apoptosis (P<0.05) and inhibited cell invasion (P<0.05). The results of western blot showed that the protein expression level of Bcl-2 was significantly decreased after treatment with glycyrrhizin (P<0.05), and the protein expression of Bax and p-Capase-3 was significantly increased (P<0.05). Conclusion: Glycyrrhizin can inhibit the proliferation and invasion and promote cell apoptosis of human tongue cancer Cal-27 cell line, which may be achieved by up-regulating the expression of Bax and p-Capase-3 and down-regulating the expression of Bcl-2.
