人绒毛膜促性腺激素对乳腺癌大鼠肿瘤生长及相关基因表达的影响

摘要:

目的:探讨人绒毛膜促性腺激素(human chorionic gonadotropin,hCG)对乳腺癌大鼠肿瘤生长及相关基因表达的影响。方法:将未经生产的40只雌性SD大鼠随机分为对照组和hCG干预组,每组20只。观察两组大鼠乳腺肿瘤的发生与生长情况,采用荧光定量PCR技术检测大鼠乳腺癌细胞以及其他正常乳腺组织中与乳腺癌相关基因的表达情况。结果:对照组大鼠的致癌例数、肿瘤数量显著高于hCG干预组(P<0.05),潜伏期短于hCG干预组(P<0.05)。hCG干预组大鼠MMP1、MMP2、MMP9、VEGF、IGF1基因表达低于对照组(P<0.05),TIMP1、PTEN、IGEIR基因表达高于对照组(P<0.05)。结论:hCG对乳腺癌大鼠的肿瘤生长起到抑制作用,对TIMP1、PTEN、IGF1R基因的表达起促进作用,对MMP1、MMP2、MMP9、VEGF、IGF1基因的表达起到抑制作用。

Objective: To investigate the effect of human chorionic gonadotropin (hCG) on tumor growth and related gene expression in breast cancer rats. Methods: Unproduced 40 female SD rats were randomly divided into control group and human chorionic gonadotropin intervention group,20 in each. The occurrence and growth of breast tumors in the two groups of rats were observed. Fluorescence quantitative PCR was used to observe the expression of genes associated with breast cancer in rat breast cancer cells and other normal breast tissues. Results: The number of carcinogens and the number of tumors in the control group were significantly higher than that in the hCG intervention group (P<0.05), and the latency time was longer than that of the hCG intervention group (P<0.05). The expressions of MMP1, MMP2, MMP9 and vascular endothelial growth factor(VEGF)in the hCG intervention group were lower than those of the control group (P<0.05), and the expression of the TIMP1 gene was higher than that of the control group (P<0.05). The expression of PTEN and IGF1R genes in the hCG intervention group was significantly higher than that in the control group, and the expression of IGF1 gene was lower than that in the control group (P<0.05). Conclusion: hCG inhibits the growth of breast cancer in rats and promotes the expression of TIMP1, PTEN and IGF1R, and inhibits the expression of RNA as well as IGF1, such as MMP1, MMP2, MMP9, VEGF and so on.
