Objective: To investigate the effect of S100A8 and S100A9 on matrix metalloproteinase (MMP) in nasopharyngeal carcinoma CNE-2 cells. Methods: Recombinant S100A8 and S100A9 siRNA vectors,pEGFP-N1-S100A8 and pEGFP-N1-S100A9 overexpression vectors were constructed and transfected into nasopharyngeal carcinoma CNE-2 cells.The transfection efficiency of each group was observed.The gene and protein expression levels of MMP7, MMP9 and MMP12 in each group were detected.The results of S100A8 siRNA group, S100A9 siRNA group,S100A8 overexpression group,S100A9 overexpression group were compared with the siRNA control group, overexpression control group and normal control group. Results: The transfection efficiency was 60%-70%. The expression levels of MMP7, MMP9 and MMP12 in S100A8-siRNA group and S100A9-siRNA group were significantly lower than those in the siRNA control group, overexpression control group and blank control group(P<0.05).The expression levels of MMP7, MMP9 and MMP12 in S100A8 overexpression group and S100A9 overexpression group were significantly higher than those in the control groups(P<0.05). Conclusion: The high expression of S100A8 and S100A9 may promote the expression of MMP7, MMP9 and MMP12, which may be related to the invasion and metastasis of nasopharyngeal carcinoma. |
