薤白皂苷对冠心病大鼠血小板聚集及凝血功能的影响

摘要:

目的:研究薤白皂苷对冠心病大鼠血小板聚集和凝血功能的影响。方法:以高脂饲料喂养及注射脑垂体后叶素建立冠心病大鼠模型,将模型大鼠随机分为薤白皂苷3个剂量组和阿司匹林组、空白对照组共5组,分别使用168、337、674 mg·kg-1薤白皂苷和阿司匹林、生理盐水进行干预。采集大鼠动脉血,制备富血小板血浆检测血小板聚集率,同时检测凝血酶原时间(PT)、凝血酶时间(TT)、活化部分凝血活酶时间(APTT)、血浆纤维蛋白原(Fib)含量。结果:离体试验中,中、高剂量薤白皂苷组血小板聚集率低于阿司匹林组(P<0.01或P<0.05)。体内试验中,高剂量薤白皂苷组血小板聚集率与阿司匹林组差异无统计学意义(P>0.05)。不同剂量的薤白皂苷及阿司匹林均能延长大鼠TT、PT、APTT。高剂量的薤白皂苷对大鼠APTT的延迟作用与阿司匹林组差异无统计学意义(P>0.05)。不同剂量的薤白皂苷及阿司匹林均明显降低大鼠血浆Fib水平,高剂量薤白皂苷组大鼠血浆Fib水平与阿司匹林组差异无统计学意义(P>0.05)。结论:薤白皂苷具有抑制冠心病模型大鼠血小板聚集作用,同时能延长大鼠动脉血的TT、PT、APTT,降低血浆Fib含量。

Objective:To study the effects of furostanol saponins from allium macrostemon bunge(FS-AMB) on platelet aggregation and coagulation in rats with coronary heart disease. Methods:Rat models of coronary heart disease were established with high fat diet and pituitrin injection. The model rats were randomly divided into experimental groups and control group. FS-AMB with different doses (high-dose, medium dose, low-dose) and aspirin were administrated in the experimental groups; normal saline were given to the control group. The treatment course was 7 days. Arterial blood of rats was collected to extract platelet. The maximum aggregation rates of platelet were detected by bioluminescence agglutination tester, and prothrombin time(PT), thrombin time(TT), activated partial thromboplastin time(APTT), and fibrinogen(Fib) content of plasma were detected by semi-automatic coagulation factor analyzer. Results:In vitro, the platelet aggregation rates of 337 and 674 mg·kg-1 FS-AMB group were lower than that of aspirin group(P<0.01 or P<0.05). In vivo, the inhibition rate of ADP-induced platelet aggregation with high dose of FS-AMB was comparable to that of aspirin(P>0.05). Different doses of FS-AMB and aspirin could prolong TT, PT and APTT. There was no significant difference in the delayed effect on ATPP between high-dose FS-AMB and aspirin(P>0.05). Different doses of FS-AMB and aspirin significantly reduced Fib levels in rats. There was no significant difference in Fib level between high-dose FS-AMB and aspirin(P>0.05). Conclusion:FS-AMB has an inhibitory effect on platelet aggregation. Besides, it can also prolong TT, PT and APTT and decrease the level of fib in plasma.
